Enzyme Mechanism

The Enzyme Mechanism software provides for rapid and easy characterization of a wide variety of enzyme reactions with just the click of a mouse button. DU® Series 600 and 7000 Spectrophotometers with this option will calculate Km, Vmax, kcat and Ki as well as the Hill constant. Up to four data plots can be displayed simultaneously (Michaelis-Menten, Lineweaver-Burk, Eadie-Hofstee, Hanes-Woolf, Hill and Inhibitor plots) without the need for a computer or external software packages.

The use of this software allows easy determination and/or confirmation of competitive, non-competitive or uncompetitive enzyme reaction mechanisms.

With the DU Series 7000, Enzyme Mechanism is included in the Kinetics Plus Software.

Features

  • Automatic blank subtraction and background correction for up to 24 blanks and samples, at user specified cell positions in the 6 or 12 position cell holder.
  • Blanks may be viewed or selected for subtraction.
  • Reaction rates can be determined automatically by the instrument or from user selected time points.
  • Choice of rate calculation methods: linear, auto-linear, first order, and auto-first order.
  • Delta A/minute, concentration and enzyme activity calculations.
  • Km, Vmax, kcat, and Ki determinations by Marquardt nonlinear regression analysis.
  • Hill constant calculation.
  • Plotting functions include split screen and overlay, zoom, curve offset, annotate and trace features.
  • Data may be averaged from 0.05 to 99.9 seconds (DU Series 600) or 0.10 to 99.9 seconds (DU Series 7000).
  • Calculation of Ki for competitive, uncompetitive and noncompetitive mechanisms.
  • Plots generated include:

    • Lineweaver-Burk
    • Eadie-Hofstee
    • Hanes-Woolf
    • Inhibition
    • Hill
  • Sample directly to and from 96-well plates with just 100 µL of sample.
  • Interval time between successive readings on a single sample may be set from 0.1 to 9999.9 seconds.
  • Selectable read average time to improve single-to-noise ratio.
  • Rates and concentrations may be entered from external studies to take advantage of the Enzyme Mechanism Software's capabilities.
  • Data from up to 10 sets of 24 samples may be accumulated and analyzed.
  • Raw and/or calculated data may be output to external devices.
  • Method parameters and sample data can be stored in separate files in the instrument for easy recall and use.
  • Cell match function provides automated zero correction of multiple cells.

Accessories
The Enzyme Mechanism Software is most efficiently operated with the Auto 6 Cell Holder or the Micro-Auto 12 Cell Holder which can be used to obtain data automatically from up to 12 samples of just 100 µL each.
