Sample Preparation Guide for Biological Tissues

Homogenates

  1. Add 0.2 mL1 of up to 10% tissue homogenate (in either water or 70% ethanol) to 1 mL of water.
  2. Add 10 mL of Ready Gel™ or Ready Safe™, shake vigorously and count.

Notes:

  1. Homogenates of up to 1 mL can be processed by following the procedure for coarse ground tissue below.

Expected tritium efficiency is 35-40%.

For homogenate assays that include a filtration step on glass fiber filters see Part 4 - Inert Supports.

Coarse Ground Tissue

  1. Add 150 mg of coarse ground tissue to 2 mL of BTS-450 and swirl.
  2. Incubate 3-5 hours at 40ºC.
  3. Add 10-15 mL of Ready Organic™ containing 7 mL glacial acetic acid per liter of cocktail1.
  4. Shake and count.

Note:

Organs

  1. Add 1 mL BTS-450 to the tissue weights shown in the following table:

      Cartilage 20- 55 mg    Nerve Cells 50-100 mg 
      Cornea 40-160 mg    Pancreas 50-140 mg 
      Heart 100-120 mg    Spleen 50-140 mg 
      Intestine 80-100 mg    Stomach 80-100 mg 
      Kidney 80-100 mg 
  2. Incubate 2-4 hours at 40ºC or until solubilized.

  3. Add 10-15 mL of Ready Organic containing 7 mL glacial acetic acid per liter of cocktail1.

  4. Shake and count.
