By taking full advantage of our expertise in both capillary electrophoresis and automation technologies, the ProteomeLab PA 800 delivers a comprehensive in-solution protein characterization (protein electrophoresis) system designed to optimize proteomics discovery.

The ProteomeLab PA 800 resolves and quantifies proteins by their isoelectric point and molecular weight, generates high-resolution peptide maps and carbohydrate profiles and provides front-end separation to mass spectrometry.


Product Features

To ensure compatibility with automated fractionation processes, sampling formats include 96-well plates, microfuge tubes, PCR* vials and 2 ml vials.

From peptides to proteins to carbohydrates, modular photodiode array and laser-induced fluorescence detection deliver high-sensitivity, quantitative analysis capabilities.

For structural elucidation, the ProteomeLab PA 800's external detector adapter allows for easy interfacing of the separation capillary with electrospray mass spectrometry.
  Temperature control of the sample compartment ranges from 5 - 60° C, maintaining protein stability while enabling temperature impact studies on enzymatic activity and protein interaction measurements.

Applications

Protein Sizing by Automated SDS-Gel Electrophoreses

SDS-PAGE is one of the most common methods used to assess the molecular weight of proteins. The ProteomeLab PA 800 automates the process of molecular weight-based purity determinations, and with on-line UV detection provides accurate quantitation without the need for staining. This means that results are obtained quickly, with no gel preparation required. Samples are treated in the same manner as SDS-PAGE (heat denaturation in the presence of SDS with optional disulfide bond reduction). And the separation is by capillary gel electrophoresis, using a formulation of polymers optimized for the resolution of proteins with a wide molecular weight range.

Figure A illustrates the analysis of size standards. Figure B highlights the analysis of a reduced IgG monoclonal antibody (L=light chain, H=heavy chain and NG=non-glycosylated heavy chain). The inset electropherogram (Figure B) highlights a non-glycosal heavy chain region.

To perform both reduced and non-reduced evaluations of immunoglobulins, the ProteomeLab SDS-MW gel analysis has been developed into an IgG assay kit. Gel buffer, sample buffer and control reagents are also available as standalone components for designing custom analyses of all other proteins.

Carbohydrate Profiling and Analysis

The ProteomeLab Carbohydrate Labeling and Analysis Kit contains the reagents, methods and controls needed for both oligosaccharide and monosaccharide analysis.

The distribution of glycans released from a glycoprotein yields a fingerprint useful in protein identification. The ProteomeLab PA 800 provides rapid analysis of released glycans, with separation times typically less than 15 minutes. Laser-induced fluorescence provides high sensitivity, enabling you to simply "dilute and shoot" samples.

The analysis of monosaccharide composition can provide valuable information during the initial characterization of a glycoprotein. A clear advantage of our ProteomeLab PA 800 approach is in the specificity of the assay. The reductive amination and open-tube format allow this analysis to be performed directly from the glycoprotein hydrolysate--so there's no need for posthydrolysis cleanup. With LIF and APTS derivization, each sugar yields the same detector response, such that their relative quantities can be directly compared.

Resolving Proteins by Charge Characteristics

By determining a protein's isoelectric point, identity can be partially established. Heterogeneity can be assessed with quantitation of the different variants separated. Along with speed and automation advantages, the ProteomeLab PA 800 automated capillary isoelectric focusing (CIEF) offers direct quantification with on-line UV detection.

The solution-based approach of the ProteomeLab PA 800 allows easy creation of an environment to assess differences in a protein's glycosylation state. By doing this directly from the protein -- through the addition of salts like borate -- complexes can be formed which amplify the charge state of the glycovariants. This is the simplest approach to assessing the heterogeneity of a glycoprotein.

The ProteomeLab Methods Development kit provides chemistry for the analysis of proteins which are highly basic or acidic. This chemistry allows you to resolve proteins by difference in mass/charge. The kit includes buffers, controls and a specialized coated capillary to minimize surface interactions.

Automated Peptide Mapping

The ProteomeLab PA 800 is optimized for high-resolution peptide mapping, utilizing detection either by UV or through external  interface to mass spectrometry (MS). By incorporating the separation prior to MS, sensitivity is increased. Plus, enhanced information on the mobility of each peptide is obtained, which can be useful in elucidating the identity of each species. The two electropherograms below illustrate the analysis of tryptic digests Cyctochrome C from two different species. Clear differences can be identified via the PA 800 and characterized further by MS.
